6个STR基因座在广东潮汕地区汉族人群多态性研究被引量:1

目的调查6个STR基因座在中国广东潮汕地区汉族人群中的基因频率分布。方法应用四色荧光标记引物复合扩增技术对251名广东潮汕地区汉族无关个体血样的6个STR基因座进行多态性研究。结果在广东潮汕地区汉族人群中6个STR基因座个体识别概率在0.798到0.925之间,杂合度在0.645到0.765之间,多态性信息含量在0.567到0.758之间。6个STR个体识别概率基因座总值为0.999994。所有基因座经χ2检验符合Hard-Weinberg平衡。结论上述6个STR基因座在广东潮汕地区汉族人群中等位基因分布较好,个体识别率高,适合法医个体识别和亲子鉴定。

Mucus hypersecretion is a distinguishing feature of chronic inflammation diseases, such as asthma,chronic bronchitis, bronchiectasis and cystic fibrosis Mucus hypersecretion leads to impairment of mucociliary clearance, abnormal bacterial plantation, mucus plug in the airway, and dysfunction of gas exchange.5 To block this vicious cycle, chronic inflammation in the airway must be controlled and mucus hypersecretion must be reduced.

Increased expression of human calcium-activated chloride channel 1 is correlated with mucus overproduction in the airways of Chinese patients with chronic obstructive pulmonary disease被引量:8

Background Chronic obstructive pulmonary disease (COPD) is usually complicated with mucus overproduction in airway. Recently the increased expression of the human calcium-activated chloride channel 1 (CaCC1) was found to play an important role in mucus overproduction in the asthmatic airways. To investigate the relationship of CaCC1 and mucus overproduction in the airway of Chinese patients with COPD, the expressions of CaCC1, MUC5AC and mucus in bronchial tissues were examined. Methods Bronchial tissues were obtained from fiberoptic bronchoscopy and bronchial biopsy in West China Hospital from April to July in 2004. Twenty-five patients were diagnosed as the patients with COPD overproduction, and other 20 were the control subjects. The expressions of CaCC1, MUC5AC and mucin in bronchial tissues were detected by reverse transcriptase-polymerase chain reaction (RT-PCR), in situ hybridization with digoxigenin (DIG)-Iabeled RNA probe, immunohistochemical and alcian blue-periodic acid Schiff (AB-PAS) staining, respectively. Results Compared with the control group, the stronger expressions of CaCC1 were further detected throughout the bronchial tissues from patients with COPD (P〈0.01). Furthermore, the stronger expressions of the CaCC1 mRNA were related to the severity of airflow obstruction. Samples from COPD showed a stronger staining for MUC5AC than those in control subjects (P〈0.01) and AB-PAS staining revealed more mucins in COPD patients' submucosal gland comparing with that in control subjects (P〈0.01). Expression levels of the CaCC1 mRNA were respectively negatively correlated with the patients' forced expiratory volume in one second (FEV~) / forced vital capacity (FVC) data, FEV1% predicted data, V50% predicted data, V25% predicted data (r=-0.43, r=-0.43, r=-0.35, r=-0.36, P〈0.01, P〈0.01, P〈0.05, P〈0.05). 汉、藏族肺结核患者血清差异蛋白质的质谱分析被引量:4

目的用表面增强激光解析电离-飞行时间质谱(SELDI-TOF MS)技术比较汉、藏族肺结核患者之间及其与相应的非结核对照者之间的血清差异蛋白,以寻找各自的质谱诊断模型并比较两个民族肺结核的蛋白质差异。方法收集2008年6月至2009年3月四川大学华西医院呼吸科结核病区、西藏自治区驻成都办事处医院内科、成都市传染病医院和成都市结核病防治研究院内科住院患者及门诊健康体检者共154份血清样本,其中87例活动性肺结核患者(汉族38例,藏族49例),67例非结核对照(汉族36例,藏族31例)。SELDI-TOF MS质谱分析分为训练组和测试组:训练组包含53例肺结核(汉族23例,藏族30例)和42例非结核对照(汉族20例,藏族22例),所获得的血清质谱用决策树的学习算法来进行分析,得到的诊断模型再用另一个独立的测试组来进行准确性验证,测试组包含59份血清样本:34例肺结核(汉族15例,藏族19例),25例非结核对照(汉族16例,藏族9例)。结果训练组中得到的诊断模型质荷比(m/z)为3193.61、4592.11可以诊断出20/23例汉族肺结核和17/20例汉族对照,准确度86.04%(敏感性为86.95%,特异性为85.00%),模型m/z为4821.45能够诊断出28/30例藏族肺结核和19/22例藏族对照,准确度90.38%(敏感性93.33%,特异性86.36%),而模型m/z为4091.98、3398.27、7970.44、4965.51、7970.44则能够将藏族肺结核从汉族患者中区分出来(准确度92.45%)。在相应的验证组中,3个诊断模型的准确度分别为70.96%、71.42%和67.92%。同时发现藏、汉族肺结核之间有16个差异蛋白峰。结论SELDI-TOF MS对汉、藏族肺结核的诊断均具有较高价值,两个民族的肺结核患者存在一定的蛋白质表达差异。

利用毛细管电泳技术分析云南回族人群7个STR基因座的遗传多态性被引量:3

目的对云南回族群体的D3S1358/THO1/D21S11/D18S51/D5S818/D13S317/D7S820等7个STR基因座的遗传多态性进行群体遗传学研究。方法利用荧光标记复合扩增及毛细管电泳自动荧光检测的方法,对204例无关个体的7个STR基因座进行检测。结果获得7个基因座的各基因座等位基因的分布频率和基因型频率,结果均符合Hardy-Weinberg平衡。计算了各基因座的杂合度(He)、个人识别能力(PD)、偶合率(PM)、非父排除率(PE)和多态性信息总量(PIC)等群体遗传学数据。结论利用毛细管电泳技术可对这7个STR基因座进行很好的检测,这些基因座多态性丰富、灵敏度高,可用于人类遗传分析及法医学中的亲子鉴定和个人识别。

Simvastatin attenuates lipopolysaccharide-induced airway mucus hypersecretion in rats被引量:12

Background Mucus hypersecretion in the respiratory tract and goblet cell metaplasia in the airway epithelium contribute to the morbidity and mortality associated with airway inflammatory diseases. This study aimed to examine the effect and mechanisms of simvastatin on airway mucus hypersecretion in rats treated with lipopolysaccharide (LPS). Methods Mucus hypersecretion in rat airways was induced by intra-tracheal instillation of LPS. Rats treated with or without LPS were administered intra-peritoneally simvastatin (5 and 20 mg/kg) for 4 days. Expression of Muc5ac, RhoA and mitogen-activated protein kinases (MAPK) p38 in lung were detected by real-time polymerase chain reaction (PCR), immunohistochemistry or Western blotting. Tumor necrosis factor (TNF)-α and IL-8 in bronchoalveolar lavage fluid (BALF) were assayed by an enzyme-linked lectin assay and enzyme linked immunosorbent assay (ELISA). Results Simvastatin attenuated LPS-induced goblet cell hyperplasia in bronchial epithelium and Muc5ac hypersecretion at both the gene and protein levels in lung (P 〈0.05). Moreover, simvastatin inhibited neutrophil accumulation and the increased concentration of TNF-α and IL-8 in BALF follows LPS stimulation (P 〈0.05). The higher dose of simvastatin was associated with a more significant reduction in Muc5ac mRNA expression, neutrophil accumulation and inflammatory cytokine release. Simultaneously, the increased expression of RhoA and p38 MAPK were observed in LPS-treated lung (P 〈0.05). Simvastatin inhibited the expression of RhoA and p38 phosphorylation in lung following LPS stimulation (P 〈0.05). However, the increased expression of p38 protein in LPS-treated lung was not affected by simvastatin administration. Conclusions Simvastatin attenuates airway mucus hypersecretion and pulmonary inflammatory damage induced by LPS. Effects of single nucleotide polymorphisms 869 T/C and 915 G/C in the exon 1 locus of transforming growth factor-β1 gene on chronic obstructive pulmonary disease susceptibility in Chinese被引量:9

Background The main risk factor for chronic obstructive pulmonary disease (COPD) is cigarette smoking. However, only 10%-20% of chronic heavy smokers develop systematic COPD. We hypothesized that the inheritance of gene polymorphisms could influence the development of COPD, which was investigated by studying two single nucleotide polymorphisms (SNP) in exon 1 of the transforming growth factor-β1 (TGF-β1) gene. Methods We enrolled 219 patients with COPD as the research group and 148 healthy people as the control group, all of whom were Chinese Han people. The polymorphisms of the TGF-β1 gene, 869T/C and 915G/C, were analyzed using the method of amplification refractory mutation system-polymerase chain reaction (ARMS-PCR). Results The occurrence of the TGF-β1 gene 869T/C polymorphism in patients with COPD was significantly different from the control group (P 〈0.05), in which the relative risk of this disease increased in cases who had the C allele (OR: 1.131, 95% CI: 1.101-1.539). There was no increased frequency of TGF-β1 915G/C gene in COPD patients compared with control subjects (P 〉0.05). Conclusions The polymorphism 869T/C in TGF-β1 gene has a significant association with disease occurrence in COPD patients and the C allele might be a risk factor. The homozygous wild-type CC of 869T/C on TGFβ1 could be a predisposing factor in COPD and those who carry the C allele might have particularly susceptibility to developing COPD.
