可溶性鸡蛋壳膜蛋白-聚丁二酸丁二醇酯共同静电纺丝负载凝血酶制备新型止血敷料的可行性研究被引量:1

目的探讨以可溶性鸡蛋壳膜蛋白(soluble eggshell membrane protein,SEP)和聚丁二酸丁二醇酯(poly butylene succinate,PBS)共同静电纺丝负载凝血酶制备新型止血敷料的可行性和有效性。方法以PBS/SEP制备新型止血敷料,通过大鼠肝脏创伤模型止血实验、髂动脉出血、肝脏止血材料植入和犬腹主动脉切开缝合后吻合口渗血模型止血实验评估其止血性能及生物安全性;选用人胚纤维母细胞,通过四唑盐比色法试验对其细胞体外毒性进行评价。结果PBS/SEP在大鼠肝脏创伤模型的止血时间和髂动脉出血实验的效果优于对照组,但犬腹主动脉渗血模型的止血时间略长,差异有统计学意义(P<0.05)。细胞毒性检测发现,不同浓度PBS/SEP浸提液在不同时间段的细胞毒性分级均为0~1级。体内实验提示,植入大鼠肝脏后可能部分影响肝功能,对机体肾功能无损害,且不引起机体全身免疫和炎症反应。结论PBS/SEP共同静电纺丝负载凝血酶止血材料是一种有效的止血材料,生物相容性良好,有良好的临床应用前景。

Improved Mechanical Properties of Poly(butylene succinate) Membrane by Co-electrospinning with Gelatin被引量:2

Gelatin, a natural proteinous polymer, was used to co-electrospin with poly(butylene succinate)(PBS) in order to improve the mechanical properties of PBS membrane and facilitate its applications in biomedical field. The PBS/gelatin blend membranes have narrower distribution of fiber diameter and smoother surface than neat PBS membrane. The contact angles, water absorption rates and water uptakes of the PBS/gelatin blend membranes were measured, showing increased hydrophilicity. The interaction between PBS and gelatin was investigated by attenuated total reflectance Fourier transform infrared spectroscopy(ATR-FTIR) and differential scanning calorimetry(DSC). The mechanical properties of PBS/gelatin blend membranes in both dry and wet states were evaluated by uniaxial tensile tests. In the dry state, the PBS/gelatin blend membrane containing 10% gelatin has a 3-times increase in tensile strength without any adverse effect on ductility because of the existence of interaction between the two blend components, little change in crystallinity of PBS, and possible interaction between any adjacent fibers; the tensile strength and elongation at break are even better in the wet state attributed to some gelatin on fiber surfaces, which act as a binder in the presence of water. The potential applications of PBS/gelatin blend membranes were demonstrated by successful immobilization of thrombin, a clinically-used hemostatic drug. The thrombin-loaded membrane could be used for rapid hemostasis.

Antibacterial Thermoplastic Polyurethane Electrospun Fiber Mats Prepared by 3-Aminopropyltriethoxysilane-assisted Adsorption of Ag Nanoparticles被引量:4

Antibacterial thermoplastic polyurethane(TPU) electrospun fiber mats were prepared by adsorption of Ag nanoparticles(Ag NPs) onto TPU/3-aminopropyltriethoxysilane(APS) co-electrospun fiber mats from silver sol. The use of APS can functionalize TPU fibers with amino groups, facilitating the adsorption of Ag NPs. The effects of p H of silver sol and APS content on Ag NP adsorption and antibacterial activity were investigated. Ag NP adsorption was evidenced by TEM, XPS and TGA. Significant Ag NP adsorption occurred at p H = 3-5. The main driving force for Ag NP adsorption is electrostatic interaction between ―NH3^+ of the fibers and ―COO-derived from the ―COOH group capped on the surfaces of Ag NPs. The antibacterial activity of the Ag NP-decorated TPU/APS fiber mats was investigated using both gram-negative Escherichia coli and gram-positive Bacillus subtilis. The antibacterial rate increases with increasing APS content up to 5% where the antibacterial rates against both types of bacteria are over 99.9%.

氨基官能化聚乳酸电纺纤维的制备、表征和应用被引量:3

通过聚乳酸(PLA)和氨基丙基三乙氧基硅烷(KH550)混合进行静电纺丝制备氨基官能化聚乳酸纳米纤维。采用滴定法测定了纤维表面氨基含量,证明当 KH550的添加量为3%~13%(质量分数)时,有19%~26%的氨基出现在纤维的表面。利用场发射扫描电子显微镜、差示扫描量热(DSC)仪、接触角测试仪和电子拉伸机对纤维形貌、 PLA 的玻璃化转变温度和熔点以及纤维膜的亲水性和力学性能进行了表征。结果表明, KH550的加入可以在电纺纤维表面引入氨基,同时使纤维直径变细,使 PLA 的玻璃化转变温度上升,熔点下降,电纺纤维膜的亲水性略有增加,力学性能有所下降。通过吸附将金纳米粒子负载到氨基官能化聚乳酸电纺纤维膜上,得到负载型催化剂,对硼氢化钠还原对硝基苯酚的反应具有良好的催化活性和重复使用性。

腹主动脉瘤腔内修复术在高龄患者中应用的研究进展被引量:4

腹主动脉瘤(abdominal aortic aneurysm,AAA)是最常见的老年主动脉疾病。流行病学研究显示,AAA在男性中的发病率为5%,但在80岁及以上患者中,发病率增长到10%,而且其瘤体破裂的风险较其他人群更高。据预测,到2020年,我国80岁以上人口将达到3000万,90岁以上的将超过1200万,在未来将有越来越多的高龄AAA患者需要接受治疗。
