温度周期性波动对凡纳滨对虾稚虾蜕壳和蜕壳激素含量的影响被引量:2

采用实验生态学方法,以温度25℃为对照,研究了四种温度波动幅度(25±1、25±2、25±3、25±4℃)对凡纳滨对虾(Litopenaeus vannamei)稚虾蜕壳率和蜕壳激素滴度的影响。主要实验结果如下:(1)25±3℃和25±4℃组对虾的蜕壳率显著高于其他各处理组(P<0.05)。(2)5种温度处理下,对虾血淋巴中蜕壳激素含量在一个蜕壳周期内的变化呈相似的规律:蜕壳间期,对虾血淋巴中蜕壳激素含量逐步增大,至蜕壳前期时,蜕壳激素的含量增至最高值;对虾蜕壳后,血淋巴中蜕壳激素含量急剧下降到一个较低的水平。(3)不同蜕壳时期的对虾在5种温度处理下,其血淋巴中蜕壳激素含量存在显著性差异;蜕壳前期时,25±3℃组对虾蜕壳间期蜕壳激素含量最高,显著高于其他各处理组(P<0.05)。实验结果初步表明,25±3℃变温处理下,蜕壳前期对虾蜕壳激素含量的增加可能是其蜕壳生长的主要原因。

光照对中国对虾稚虾3种消化酶活力的影响被引量:14

在实验室条件下研究光照(光照周期、光照强度和光色)对中国对虾(Fenneropenaeus chinensis)稚虾3种消化酶(蛋白酶、淀粉酶和脂肪酶)活力的影响。在本实验条件下,主要结果如下:(1)4种光照周期下,对虾3种消化酶的活力差异不显著(P>0.05);(2)在完全黑暗、6μmol.m-2.s-1、30μmol.m-2.s-1和110μmol.m-2.s-1光照强度下,对虾蛋白酶的活力分别为0.361 U.mg-1(protein)、0.081 U.mg-1(protein)、0.088 U.mg-1(protein)和0.273 U.mg-1(protein),完全黑暗和110μmol.m-2.s-1下的活力显著高于其他两个光照强度(P<0.05),且两者间的差异达到显著水平(P<0.05);中国对虾淀粉酶的活力分别为0.386 U.mg-1(protein)、0.095 U.mg-1(protein)、0.111 U.mg-1(protein)和0.315 U.mg-1(protein),完全黑暗和110μmol.m-2.s-1条件下的活力显著高于其他两个光照强度(P<0.05),两者间的差异亦达到显著水平(P<0.05);对虾脂肪酶的活力分别为0.147×10-2U.mg-1(protein)、0.150×10-2U.mg-1(protein)、0.170×10-2U.mg-1(protein)和0.183×10-2U.mg-1(protein),完全黑暗和110μmol.m-2.s-1条件下的活力差异达到显著水平(P<0.05);(3)不同光色下,对虾蛋白酶和淀粉酶的活力差异不显著(P>0.05),脂肪酶的活力则存在一定的差异,蓝光和黄光下,对虾脂肪酶的活力分别为0.093×10-2U.mg-1(protein)和0.107×10-2U.mg-1(protein),显著高于白光和绿光下的活力(P<0.05),但蓝光组和黄光组间的差异不显著(P>0.05)。

盐度和Ca^(2+)浓度对中国明对虾稚虾耗氧率的影响被引量:6

设计双因子实验研究了水温25.0℃±0.5℃下,盐度(5,15,30)和Ca2+质量浓度(175,350,700,1 400,2 800 mg/L)对中国明对虾(Fenneropenaeus chinensis)稚虾耗氧率的影响,实验对虾的湿体质量为0.301 g±0.041 g。实验结果表明:(1)不同盐度下,中国明对虾稚虾耗氧率的大小顺序为R5>R30>R15。其中,盐度15下对虾的耗氧率显著低于盐度5和30下的耗氧率(P<0.05);(2)不同Ca2+质量浓度下,中国明对虾稚虾耗氧率的大小顺序为R2 800>R175>R700>R1 400>R350。其中,Ca2+质量浓度为350 mg/L组对虾的耗氧率显著低于其它处理组,Ca2+质量浓度为2 800 mg/L组对虾的耗氧率显著高于其它处理组(P<0.05),而Ca2+质量浓度为175,700和1 400 mg/L组间对虾的耗氧率差异不显著(P>0.05);(3)盐度和Ca2+质量浓度的交互作用显著影响中国明对虾稚虾的耗氧率(P<0.05)。

研究不同频率的盐度波动模式下,中国明对虾稚虾的蜕皮、生长和能量收支。实验结果表明:(1)盐度波动D4(对虾在高盐度水平下处理4 d,降至低盐度水平处理2 d后回到高盐度水平,依次往复,D4表示在高盐水平停留4 d,其它略同)和D6组对虾的蜕皮率分别比恒定盐度D0组高12.9%和17.4%,且D6组和D0组间对虾的蜕皮率差异达到显著水平(P<0.05);(2)盐度波动D4组对虾的特定生长率显著高于其它4个盐度处理组(P<0.05);(3)盐度波动组和恒定盐度组间对虾的摄食量差异不显著(P>0.05);(4)盐度波动D4组对虾的食物转化率显著高于恒定盐度D0组26.9%(P<0.05);(5)盐度波动D4组对虾用在生长的能量比例显著高于恒定盐度D0组25.6%,(P<0.05);对虾用在蜕皮上的能量比例以D6组最高,D4组次之,且D6组与D0组间的差异达到显著水平(P<0.05);对虾用在呼吸和排泄上的能量比例各处理组间存在一定的差异(P<0.05),而用在排粪上的能量比例差异则不显著(P>0.05)。D6组对虾较高的蜕皮能表现出较高的蜕皮率;D4组对虾较高的生长能是其生长较快的原因。

在实验室条件下,研究了不同Ca2+浓度波动幅度对凡纳滨对虾(Litopenaeus vannamei)稚虾蜕皮、生长和能量收支的影响。实验在水族箱内进行,实验对虾的初始体质量为(1.459±0.006)g,实验用水为人工海水,盐度为30,水温(25.0±0.5)℃,实验设恒定Ca2+质量浓度360mg/L为对照组(C360),4个实验组的Ca2+质量浓度波动幅度分别为60mg/L(C300)、120mg/L(C240)、180mg/L(C180)和240mg/L(C120),波动周期为4d。实验持续40d,主要实验结果如下:(1)不同Ca2+浓度处理下,对虾的蜕皮率存在一定的差异。其中,C120组对虾的蜕皮率最高,达(11.0±0.5)%/d,显著高于其他处理组(P<0.05),而其他处理组之间差异不显著(P>0.05)。(2)不同Ca2+浓度处理下,对虾的特定生长率(SGR)由大到小依次为C360、C300、C240、C180、C120。其中,C120组对虾的特定生长率显著低于其他处理组(P<0.05),而其他处理组之间的差异未达到显著水平(P>0.05)。(3)不同Ca2+浓度处理下,对虾的摄食率(FI)差异不显著,而对虾的食物转化效率(FCE)存在一定的差异。其中,C120组对虾的食物转化效率最低,显著低于其他处理组(P<0.05),而其他处理组之间差异不显著(P>0.05)。(4)不同Ca2+浓度波动对凡纳滨对虾稚虾的生长能、排粪能和蜕壳能占摄食能的比例影响显著(P<0.05)。实验结果表明,水中Ca2+浓度波动幅度过大会刺激对虾蜕皮,一定程度上影响对虾的生长。

采用实验生态学方法研究了在盐度为20的条件下,4个盐度波动幅度(2、4、6、8)对中国对虾稚虾蜕皮和生长的影响.结果表明:中国对虾的蜕皮率为13.3%~15.4%,处理间差异未达到显著水平;盐度波动幅度为4的情况下,对虾的特定生长率最大,用在生长上的能量最高,用于呼吸的能量最低,生长迅速;盐度波动幅度为2的情况下,中国对虾的摄食量最低;盐度波动幅度为2和4的情况下,对虾的食物转化效率最高;不同处理中国对虾用在蜕皮上的能量差异不显著.盐度波动幅度过大不利于中国对虾稚虾的生长,但对蜕皮无影响.

探讨不同的pH变动模式对对虾生长的影响,设计2个独立的实验,分别研究pH值为8.9和4.0的不同时间的处理条件下,酸碱度的周期性变化对凡纳滨对虾的生长的影响,实验1设置了2个恒定pH组(7.9±0.15和4.0±0.10)和5个pH周期性变动的处理:对虾每2天在pH=(4.0±0.10)下分别处理0.5,1,2,4或12 h,其它时间处在正常pH=7.9中。结果表明,每2天1 h以上的pH=4.0的处理就能显著阻碍对虾的生长,而每2天12 h以上的pH=4.0的处理就能显著影响对虾的成活率。实验2设置了两个恒定pH组(分别为7.9±0.15和8.9±0.10)和6个pH周期性变动的处理:对虾每天在pH=8.9±0.10下分别处理0.5,1,2,4,8或12 h,其它时间处在正常pH中(7.9)。结果表明恒定的高pH(8.9±0.10)显著抑制了对虾的生长,而每天给予凡纳滨对虾2 h和4 h的pH=8.9的刺激则能提高其摄食率和食物转化率,并显著促进其生长。总之,pH值周期性地向酸性环境方向波动会明显妨碍对虾的生长,而向碱性环境方向的波动则会促进对虾的生长,而且这一影响的强度与波动持续的时间长短有关。

Effects of Salinity Fluctuation Frequency on the Growth, Molting Rate and Hemolymph 20-Hydroxyecdysone Concentration in Juvenile Chinese shrimp, Fenneropenaeus chinensis被引量:3

This experiment was conducted to examine the effects of salinity fluctuation frequency on the growth, molting rate and hemolymph 20-hydroxyecdysone (20-HE) concentration for juvenile Fenneropenaeus chinensis with an initial wet body weight of 1.115 g±0.012 g. The salinity of the control treatment (represented by DO) was 28 throughout the experiment, whereas treatments D2, D4, D6 and D8 were subjected to different salinity fluctuation frequencies of 2, 4, 6 and 8 d, respectively. The range of salinity fluctuation was 4. The salinity in treatments D2, D4, D6 and D8 was kept at 28 for 2, 4, 6 and 8d, respectively; each original salinity was decreased abruptly to salinity 24, which first lasted for another 2 d and was then raised to its initial value 28. This constituted a salinity fluctuation cycle and afterwards the cycle repeated. It was found that: 1) The weight gain of shrimp in treatment D4 was 202.9% and significantly higher than that in treatment DO (P〈0.05). The molting rate in treatment D2 was the lowest, while that in treatment D6 was the highest and significantly higher than those in treatments DO and D2 (P〈0.05). 2) The hemolymph 20-HE concentration of shrimps in all treatments was at low levels and increased first gradually during the inter-molt period and then increased sharply. It reached a peak value during the pre-molt stage, and then decreased abruptly during the post-molt stage when the lowest level occurred. 3) During the post-molt stage, the hemolymph 20-HE concentration in treatment D2 was 7.47pg μL^-1 and significantly higher than that in treatment DO (P〈0.05). There were, however, no significant differences in hemolymph 20-HE concentration among all five treatments during the inter-molt stage (P〉0.05). Experiments were conducted to examine the effects of salinity fluctuation frequency on the osmolarity, Na^+-K^+-ATPase activity and HSP70 of Chinese shrimp Fenneropenaeus chinensis ruth initial wet body weight of 1.460g±0.091 g. The salinity in the control group (DO) was 28 throughout the experiment, whereas treatments D2, D4, D6 and D8 were subjected to different salinity fluctuation frequencies of 2, 4, 6 and 8d, respectively. The salinity in treatments D2, D4, D6 and D8 was kept at 28 for 2, 4, 6 and 8d, respectively, decreased abruptly to salinity 24, lasted for another 2 d, and then was raised to its initial value 28. This was a complete salinity fluctuation cycle that afterwards repeated itself. After 32 days, the osmolarity in treatments D2, D4, D6 and D8 was significantly lower than that in treatment DO (P〈0.05). There were significant differences in both muscle and eyestalks HSP70 expression among groups. The HSP70 expressions in muscle and eyestalks in group D4 were 61.4% and 57.0% higher, respectively, than that in the control group DO (P〈0.05). There were, however, no significant differences in gill or hepatopancreas Na^+-K^+-ATPase activity between the treatments and the control.

The Effects of Rapid Temperature Changes on HK,PK and HSP70 of Litopenaeus vannamei in Different Seasons被引量:4

Activities of hexokinase(HK),pyruvate kinase(PK) and levels of HSP70 were measured to evaluate the response of Litopenaeus vannamei to rapid temperature changes under controlled laboratory conditions.Shrimps were subjected to a quick temperature change from 27℃ to 17℃ for the summer case(Cold temperature treatment),or from 17℃ to 27℃ for the winter case(Warm temperature treatment).After 0.5,1,3,6,12,24,48,and 72 h of exposure time,shrimps were sampled and prepared for further analysis.The results showed that the effect of acute temperature changes on activities of HK was significant.Patterns of variations of the two glycolytic enzymes suggested that enzymes in the glycolysis cycle could adjust their activities to meet the acute temperature change.The HSP70 level increased in both cold and warm temperature treatments,suggesting that the rapid temperature changes activated the process of body's self-protection.But the difference in expression peak of HSP70 might be related to the different body size and the higher thermal sensitivity to temperature increase than to temperature decrease of L.vannamei.
