Design of multiplex logic gates:combining regulation of DNA structure with logical calculation

In this paper,we proposed a facile and accurate way for controlling multiplex fluorescent logic gates through changing the exciting and the observing wavelengths.As proof-of-principle,a Pb2+-specific DNAzyme probe and a thymine(T)-rich DNA probe were introduced to a double-stranded(ds-)DNA.The addition style of the two ions served as the four inputs by changing the distance of the three fluorophores,6-carboxyfluorescein(FAM),ALEXA 532(ALEXA)and carboxytetramethylrhodamine(TAMRA),all of which were modified on the dsDNA probe.Compared with the previous methods,the present approach needed neither different inputs nor the change of sequence of the probe to achieve multiplex logic gates.Furthermore,the modularity of the strategy may allow it to be extended to other types of logic gates.

A new colorimetric and fluorescent probe,2-(2,4-dinitrostyryl)-1,3,3-trimethyl-3H-indolium iodide (DTI),for selective and sensitive detection of biological thiols is reported.In aqueous solution at physiological pH 7.4,biological thiols react with DTI via Michael addition to give the brownish red adduct concomitant with fluorescence emission decrease.

Fluorescent detection of singlet oxygen: Amplifying signal transduction and improving sensitivity based on intramolecular FRET of anthryl appended porphyrins被引量:3

Two new anthryl-appended porphyrin dyads have been synthesized and used as highly selective and sensitive fluorescence probes for singlet oxygen ( 1O2 ). The design strategy for the probes is directed by the idea of intramolecular fluorescence resonance ener- gy transfer (FRET) interactions and carried out by incorporation of an electron-rich fluorophore (porphyrin) with a reactive anthracene for 1O2 . The molecular recognition is based on the specific interaction of 1O2 with the inner anthracene moiety, and the signal reporter for the recognition process is the porphyrin fluorescence. As a result of overlap of the emission band of the anthracene with the absorbance band of the porphyrin, intramolecular FRET occurs between the anthracene (donor) and the porphyrin (acceptor). The effective light absorbed by the porphyrin and, concomitantly, the emitted light intensity are thus modulated by the emission intensity of the anthracene. Upon reaction with reactive oxygen species such as hydrogen peroxide, hypochlorite, superoxide, hydroxyl radicals, and 1O2 , the probes exhibit a selective response toward 1O2 . In addition, significant amplification of the signal transducer is observed. The feasibility of the design was demonstrated by monitoring the 1O2 generated from a MoO42/H2O2 system. The results clearly demonstrate that the synthesized probes exhibit both high selectivity and high sensitivity for 1O2 . The fluorescence reaction and signal amplification mechanism of the system were both discussed, clearly confirming that the introduction of electron-rich porphyrin units into the 9,10-positions of anthracene can improve the response sensitivity and activate the probe's reactivity toward 1O2 .

核酸酶催化放大传感体系的设计及其在铅离子比色检测中的应用被引量:7

基于8-17 DNA酶的催化切割特性和类辣根过氧化物酶DNA酶的氧化还原反应催化特性,发展了一种新型核酸酶催化放大传感体系,并用于Pb2+的比色检测。考察了K+浓度,pH值及反应时间对检测体系的影响。ABTS吸光度变化与Pb2+浓度在5.0~100 nmol/L范围内呈良好的线性关系,检出限为3.0 nmol/L。此外,因Pb2+酶的特异性,本方法对Pb2+具有良好的选择性。

单链DNA/表面活性剂与单壁碳纳米管的亲合力及荧光检测性能(英文)被引量:1

研制一种基于单链DNA与单壁碳纳米管荧光变化的新型荧光探针用于表面活性剂物质的检测方法。考察了温度对探针稳定性的影响,通过观察实时荧光光谱变化来评估单壁碳纳米管与5种常见表面活性剂物质的亲和性,观察镁离子及pH对探针检测表面活性物质的影响,通过观察荧光发射光谱变化来考察加入不同浓度表面活性物质曲拉通X-100后荧光探针的荧光恢复情况。结果表明,在5~80℃的温度范围内,探针十分稳定;单壁碳纳米管与5种常见表面活性物质的亲和力由强至弱依次为曲拉通X.100、十二烷基苯磺酸钠、吐温-20、吐温-80、十二烷硫酸钠;荧光探针检测的最佳Mg抖浓度为10mmol/L,pH为7.4;当加入不同浓度的表面活性剂物质曲拉通X.100时,探针体系的荧光逐渐得到恢复。
