A toxicological study of inhalable particulates by plasmid DNA assay:A case study from Macao被引量:1

Oxidative damage to plasmid DNA induced by airborne PM10 (particulate matter with an aerodynamic diameter of 10 μm or less) is caused by the bioavailable (i.e., soluble) heavy metals on the particle surface. However, quantitative analyses of the links between PM10 and oxidative damage are limited. In this study, plasmid DNA assay and ICP-MS were applied to study oxidative capacity and trace element compositions, respectively, of summer and winter PM10 samples collected at several sites (Sun Yat Sen Municipal Park (SYSP) and Av. de Horta e Costa (AHC) on the Macao peninsula and Macao University on Tai- pa Island (TI)) in Macao. At AHC and TI, the oxidative capacity of PM10 collected in winter was higher than that collected in summer, for both the whole sample and the water-soluble fraction. In contrast, no seasonal variation was noted at SYSP. PMI0 exhibited the highest oxidative capacity at SYSP and lowest oxidative capacity at TI in both seasons, demonstrating that the PMl0 collected on the Macao peninsula had a higher toxicity than that from Taipa Island. ICP-MS analyses revealed that the concentrations of total analyzed trace elements and their water-soluble components in PMI0 from TI and AHC were higher in winter than in summer, whereas SYSP displayed the opposite trend. The extents of oxidative damage induced by the wa- ter-soluble fractions and intact whole particles were generally similar, implying that the oxidative damage caused by particles in Macao resulted mainly from the water-soluble fraction. The oxidative capacities of PM10 were positively correlated with both whole and soluble Zn at the 95% confidence level, indicating that Zn was the major element responsible for the oxidative damage caused by particles in Macao. Other heavy metals, such as Cr, Cu, Cd, Ni, As, and Pb, also exhibited elevated concen- trations, and the potential health impacts of these metals should be considered.

黔西织纳煤田上二叠统层序地层及聚煤作用被引量:39

贵州西部织(织金)纳(纳雍)煤田以发育海陆过渡相含煤岩系而成为研究西南地区晚二叠世沉积相及层序地层的理想地区。作者对该煤田上二叠统含煤岩系(龙潭组和长兴组)进行了高分辨率层序地层分析,并探讨了层序地层格架下煤层厚度的变化规律。以区域不整合面以及下切谷砂体底部冲刷面等为层序界面,结合石灰岩标志层向古陆方向延伸距离而显示的旋回性特征,将该煤田上二叠统含煤岩系划分为3个三级层序。其中层序Ⅲ中煤层最厚,层序Ⅰ中煤层最薄;同一个层序中,又以海侵体系域煤层最厚,低位体系域煤层最薄。在建立黔西织纳煤田层序地层格架的基础上,指出厚煤层主要发育于海侵体系域的三角洲平原环境。

澳门可吸入颗粒物的氧化性损伤能力及原因探讨被引量:8

运用质粒DNA评价法对澳门半岛的中山公园及高士德马路采样点以及氹仔岛的澳门大学采样点的冬季和夏季可吸入颗粒物(PM10)的氧化性损伤能力进行研究,同时利用电感耦合等离子质谱(ICP-MS)对可吸入颗粒物的重金属元素组成进行分析.结果表明:澳门大学和高士德马路采样点的PM10氧化性损伤能力表现为冬季>夏季,中山公园采样点显示出冬季和夏季氧化性损伤能力基本相同;而同一季节不同采样点的大气PM10的氧化性损伤能力为中山公园采样点最高,其次是高士德马路采样点,澳门大学采样点相对较低,说明澳门半岛采样点的PM10的氧化性损伤能力大于氹仔岛采样点.ICP-MS分析结果显示,全样和水溶微量元素总含量在澳门大学和高士德马路均呈现冬季>夏季,而在中山公园采样点PM10的水溶性微量元素却是冬季<夏季.总体上澳门三个采样点的PM10的全样及水溶样的氧化性损伤能力相似,说明氧化性损伤能力主要来自于水溶组分.通过微量元素含量与颗粒物水溶部分和全样DNA损伤率的对比得出,在95%置信度上,在所有PM10样品中与氧化性损伤能力相关性最明显的是Zn元素.其他的重金属元素如As,Cd,Cr,Cu,Ni和Pb等,因具有较高的含量,亦值得今后研究重视.

云南宣威地区燃煤排放PM_(10)的微观形貌和粒度特征被引量:3

宣威地区肺癌发病率居全国首位,并且肺癌与室内燃煤关系密切. 为了解宣威地区室内燃煤排放PM10的微观形貌和粒度特征,使用小流量Minivol采样器,分别于2011年1月、3月和12月对宣威地区6个代表性乡镇的18个农户及对照点--贵州六盘水幸福村2个农户室内、外的PM10进行采样. 采用扫描电子显微镜分析样品的微观形貌,并对颗粒物粒度进行统计分析. 根据微观形貌特征将PM10分为烟尘集合体(链状、蓬松状、密实状)、球形颗粒(燃煤飞灰和焦油球)、矿物颗粒(规则和不规则状)、生物颗粒及超细未知颗粒. 结果表明:宣威地区室内PM10微观形貌差别较大;室、内外PM10的粒度分布在0.1-0.4μm内的颗粒数量所占比例较大,而1μm以上颗粒体积所占比例较大;小粒径颗粒对颗粒物数量贡献较大,而少数大粒径颗粒对体积贡献较大;宣威地区和对照点室内的PM10数量-粒度和体积-粒度分布相似,室外PM10数量-粒度特征相似但体积-粒度分布有所差异. 宣威地区室内的烟尘集合体和球形颗粒分别占颗粒物总数的10.5%和23.6%,高于对照点 (分别为7.7%和11.3%).

采用质粒DNA评价法评价哈尔滨市市区、郊区和清洁对照点的PM10和PM2.5样品的生物活性及颗粒物中金属元素对质粒DNA的氧化性损伤能力。研究结果表明,水溶样和全样样品都具有一定的活性,样品的TD40值为67.9~1 570μg/mL,说明PM的生物活性主要与其来源和性质有关;颗粒物中具有生物活性的元素主要为过渡金属,其次为碱金属或其他主族元素,除过渡金属外,稀土元素对颗粒物的生物活性也具有显著影响。水溶样的TD40值与Y的浓度呈正相关,与La,Lu,Ho,Sm的浓度呈负相关,说明在对质粒DNA损伤过程中,Y拮抗La,Lu,Ho,Sm的损伤作用;全样的TD40值与元素Cs,La,Mn,Pb,Tm,Gd的浓度呈正相关,与Tb,Ni,Yb,Er,W,Cd,In的浓度呈负相关,在对质粒DNA损伤过程中,元素Cs,La,Mn,Pb,Tm,Gd协同拮抗Tb,Ni,Yb,Er,W,Cd,In的损伤作用。

使用质粒DNA评价法和ICP-MS研究贵州织金地区海陆过渡相的晚二叠世煤及大同地区陆相的侏罗纪煤在实验燃烧炉中燃烧排放PM10对质粒DNA的氧化性损伤能力及其与重金属元素组成的关系。选择2种煤的蜂窝型煤进行燃烧实验,以木炭作为对照组进行研究。对比各种燃料燃烧产生的PM10的DNA损伤实验结果发现:全样和水溶性组分所产生的DNA损伤率相似,说明水溶性组分是造成氧化性损伤的主要来源;海陆过渡相的织金煤燃烧排放PM10的氧化性损伤能力明显比陆相大同煤燃烧产生的PM10的高,2种煤的燃烧排放PM10对DNA损伤能力均比木炭本底的高。对3种PM10中的水溶性微量元素进行分析,水溶性的As,Cd,Cu,Ge,Hg,Mo,Pb,Sb,Se,V和Zn在海陆过渡相的织金煤燃烧排放的PM10中含量较高,在陆相的大同煤燃烧排放的PM10中较低,在木炭本底燃烧排放的PM10中最低。PM10中较高的水溶性Zn和Pb含量,说明环境大气PM10中的Zn和Pb可能与燃煤排放相关,而Hg元素因为较强的挥发性而在PM10中未得到富集。水溶性的微量元素总量及大部分水溶性的单个微量元素均表现出与颗粒物对DNA损伤率一致的变化规律,说明燃煤产生的PM10与其中的水溶性组分密切相关。

采用EXXON公司"Vail"学派的观点,将晚二叠世划分为2个三级层序。层序Ⅰ发育低位体系域、海侵体系域及高位体系域,比较完整。层序Ⅱ只发育海侵体系域及高位体系域。通过绘制各种单因素等值线图,并与单井剖面相和连井剖面相分析相结合,从点到线再到面,展开古地理分析。2个层序均反映出海侵自东向西的脉动推进、东深西浅的格局,从而产生了潮坪三角洲-海湾(泻湖)-潮坪-浅海沉积模式在垂向上和平面上的展布特征,其中海湾-潮坪及碳酸盐台地构成了研究区基本骨架。物源区主要来自西部康滇古陆。总体上晚二叠世研究区遭遇了一个海水自东北向西南不断侵进的过程,因此成煤环境随着海平面的上升而不断向西南迁移。煤层主要发育在海湾-潮坪环境里,其次为低地残积平原。

Sequence Stratigraphy of the Jurassic Coal Measures in Northwestern China被引量:12

The sequence stratigraphy of the Jurassic coal measures of northwestern China has been studied based on data from outcrop and borehole sections. Because of the geological background and the diversity of basin types, such as isochronisms of coal seams and recognition of key sequence boundaries, it is rare to summarize and correlate sequence structure and framework. The types and recognition characteristics of the sequence boundaries of the Jurassic coal measures are revealed by comparing climatic characteristics, structural styles and the base level cycle. A total of five third-order sequences and 15 systems tracts have been subdivided and the sequence stratigraphic framework has been reconstructed confirming that the thick coals accumulated in the late stage of transgression system tracts of sequence I (SQ I) and sequence III (SQ III). This idea is of important academic significance for instructing coal resources exploration, and enhancing geological effects of prospecting engineering. It is also of practical significance for guaranteeing construction of the large-scale coal production base in northwestern China, accelerating the westward development of the coal industry stratagem.

重庆地区上二叠统包括龙潭组/吴家坪组和长兴组,主要发育海陆过渡相(海湾-泻湖-潮坪)及浅海碳酸盐沉积,其中龙潭组和吴家坪组为同期异相沉积。通过对区内钻井剖面及露头剖面的分析,在本区上二叠统中识别出区域性构造不整合面和沉积相转换面等地层界面,并将上二叠统划分为2个三级层序。基于近海煤盆地的背景,研究区厚煤层常靠近初始海泛面发育。总体来看,近海含煤盆地三级层序以海侵体系域聚煤最好。

Lacustrine basin evolution and coal accumulation of the Middle Jurassic in the Saishiteng coalfield, northern Qaidam Basin, China被引量:2

Based on an extensive borehole survey of the Middle Jurassic coat-bearing sequences in the Saishiteng coalfield, northern Qaidam Basin (NQB), a total of 20 rock types and 5 sedimentary facies were identified, including braided river, meandering river, braided delta, meandering river delta, and lacustrine facies. The distribution of rock types and sedimentary facies contributed to the reconstruction of three periods' sedimentary facies maps of the Middle Jurassic in the Saishiteng coalfield, namely, the Dameigou age, the early Shimengou age and the late Shimengou age. That also provided the basis for the development of a three-stage depositional model of the Middle Jurassic in the NQB, indicating the lacustrine basin of the NQB in the Dameigou age and early Shimengou age were corresponding to an overfill basin, and that in the late Shimengou age was related to a balanced-fill basin. The analysis of the stability and structure of coat seams based on sedimentary facies maps showed that the preferred coal-forming facies in the Saishiteng coalfield were inter-delta bay and interdistributary bay of lower delta plain in the Dameigou age. In particular, the swamps that developed on the subaqueous pataeohigh favored the development of thick coat seams. Thus, ruinable coal seams may also be found along the Pingtai pataeohigh in the western part of the Saishiteng coalfield.
